David's recommendations:

We just used Tris Buffer at ph10 in the microwave. You will need to figure
out your own timing but the trick is to boil for 20 mins. No real trick
here it's just a matter of being able to open up those determinents. I have
not tried other methods but I'm sure other methods would work as well if
not better. When I spoke with a tech at Pharmigen thay told me they haven't
been able to get it to work on formalin fixed tissue...Hmmmmm!!! We decided
to give it a shot and from what we can tell there is some real staining.

Your welcome. I can't tell you. I sent my slides to the histochem lab for
them to run it thru their ag. ret. method since they have the timing all set
up. I cheated sorry.
Our control primarily were those cell undergoing granular vacuolar
degeneration which has been reported in literature to be associated with
Caspase 3 activity.
