Dear Linda
I believe we spoke about the presence of antigens on the surface of blood
cells and creating agglutination in a test tube as in blood bank, there are
many antibodies that hate heat, others that like cold, some we enzyme away,
some we use enhancing agents to get them to show there presence in a
stronger
way, I use this thinking in ihc somewhat , and in the way I approach certain
antibodies or groups of antibodies(ie:cytokeratins),While it is not exactly
the same , since our tissue is preserved(formalin and alcohol,et al) it does
help me to usually achieve some success when doing new antibodies.I usuallt
test my antibodies in a variety of ways, and then when similiar groups come
along I start with what worked best in the past first. Hope this is helpful,
feel free to post on histonet if you like.
Dana
