Does anyone out there have any experience staininig for Hyaluronic
Acid Binding Protein or proteoglycans in general?
I have been trying to stain (human) skin for a panel of proteoglycans
and HABP is the latest request. The protocol that I have from
Seikagaku uses a 2 hour incubation in Sigma chondroinase for
"unmasking" and digestion in hyaluronidase for the negative control.
Another journal paper that I have doesn't use the chondroitinase step
at all. I am using formalin-fixed, paraffin sections and have had
mixed results with the other proteoglycans so far.
Any tips would be greatly appreciated.
