How this is done depends on what you are wish to examine.
Are you interested in dealing with the epidermis only or with the connective
tissue? Also if dealing with the connective tissue do you also need to
include the hypodermis?
Epidermis and dermis can be separated using EDTA and other techniques and
individual cells prepared using trypsin and oher proteases depending on your
requirments.
