The partial reversal of fibre demonstration at pH4.5/4.6 is the most
difficult of the ATPase methods to get to work. Correct pHing is the
key.
Also remember we glibly talk about ATPase at various pHs, but what we
are really doing is preincubating in acid buffers, and then doing ATPase
at pH 9.4 on all the sections! A common misconception repeated in almost
every textbook.
Regards.
