Re: IHC
I don't seem to be able to get all the basal cells in prostate staining with
protease. I use tonsil/prostate combination as a control. Tonsil stains
fine, but a lot of basal cells in prostate do not stain, whereas with HIER I
get good staining. Which protease do you use and what detection system?
Maybe my detection is not sensitive enough.

"open"too many nuclei to staining , I have even forced false positivity by
steaming too long, it is a very touchy balance. however on 34be12 we use
enzyme not steam, with no problems and do the same on cytokeratins. we use a
protease for anywhere from 2 to 6 minutes and have no problems. hope this
helps. dana
