I am presently doing immunocytochemistry and in situ hybridization experiments on embryonic day 7 chicken embryos fixed in 4% paraformaldehyde (PFA), Bouin or Carnoy and embedded in paraffin . Unfortunatly, the eye of the embryos are collapsing, and the PFA fixed heads have the tendency to harden so that I cannot section the tissue without tearing it. Do you know what can cause those problems and how I can fix them?
