Hi Lin,
I got COX-2 to work in rat lungs using COX-2 (murine) Polyclonal
Ab from Cayman Chemical (Cat. # 160126). At that time I was
using citrate buffer retreival in steam for 20 mins. and overnight
incubation at 4 C. My detection system was the LSAB2 kit from
Dako.
Subsequently, in trying to get canine tissues to stain, I have
modified to no retreival and 60-90 mins at 37 C incubation. I am still
getting a good signal (I think!) but I ahve some annoying nonsp. rx in
my deletion controls that I am having trouble finding the source of.
But I think that is an issue of species, not protocol per se.
Cox-2 does seem to be a "finicky" one to get to work!
