I searched my old lab manuals and found this:
MULLIGANS METHOD FOR MUELIN
(THOMPSON)
Principle:
Not clear but myelinited areas of the brain are
stained blue.
Solutions:
A. Mulligans solution:
Phenol Crystals 5 g
Copper sulphate 0.5 g
Hydrochloric Acid 0.125 ml
Distilled Water 100 ml
B. 2% Aqueous Ferric Chloride
C. 1% Potassium Ferrocyanide
D. Acidic Formalin:
Formalin 10 ml
Citric Acid 0.2 g
Distilled Water 90 ml
E. Mounting Fluid:
Glycerine 25 ml
Distilled Water 75 ml
Formaldehyde 10 ml
Citric Acid 0.2 g
Method:
1. Ensure that brain slices are well fixed in 10%
formalin.
2. Wash in running water, 1 hour.
3. Place for 5 minutes in solution A at 60oC.
4. Transfer to cold water, 10 seconds.
5. Transfer to solution B, 1 minute.
6. Wash in running water, 2 minutes.
7. Transfer to solution C, 4 minutes.
8. Wash in running water overnight.
9. Store in solution D for 2 months.
10. Mount in solution E.
Note: These times are precise, especially in
solution A. Otherwise, white matter will take up the
stain.
