Epitope
This antibody's target epitope is distinct from those of other anti-CD34
reactive mabs as demonstrated by its lack of sensitivity to neuraminidase
treatment, and fluorescence staining profiles during flow cytometry analysis
of target cell populations.
Specificity
This antibody recognizes the CD34 antigen, some cross reactions with
vascular associated adventitia and some basement membranes.
Ultrastructure studies, using an indirect immuno- gold technique on ultra
thin sections of human umbilical artery, have revealed that this antigen is
associated to a degree with the basoluminal surfaces of endothelium and
shows a slight reaction with an intracytoplasmic vesicular compartment.
Predominant staining is on the endothelial cell membrane.
QBEND-10 immunoprecipitates a glycoprotein with a relative MW 110kD
expressed on haematopoietic cells and on the established myeloid leukaemic
cell line KG1A.

Uses
1) Suitable for immunohistochemical staining of frozen tissues sections post
fixed with formalin, acetone chloroform-acetone 1:1 at -30°C, glutaraldehyde
and methacarn; or formalin fixed paraffin embedded material.
The antigen is labile to ethanol and methanol, with staining being greatly
reduced or lost
2) Applications for haematology- bone marrow transplantation:
Functional assays for specificity against haemapoietic progentior cells were
demonstrated in preliminary clinical studies by its effect on in vitro bone
marrow colony formation.
Depletion of CD34(+) bone marrow cells, with this antibody substantially
inhibited the growth of colonies.
Conversely, enrichment of CD34(+) cells, significantly increased the number
of colonies formed by myeloid progenitor cells. QBEND-10 is currently being
evaluated by clinical haematologists for its efficiency in ex vivo
enrichment of haemapoietic stem cells for bone marrow transplantation.
QBEND-10 was found to selectively label CD34(+) peripheral blood mononuclear
cells during the period of vigorous haemopoietic recovery, in patients
undergoing chemotherapy, by flow cytometry analysis.
Working dilution for flow cytometry 10 µl for 106 cells, a dilution of
approximately 1:5 is advised.
For immunohistochemistry 1:10 dilution is suggested.
Applicable working dilutions for immunoprecipitation and Western blotting
should be tested on each individual laboratory.
Positive control
Aceton fixed rat lung endothelial cell (90% RECA positive) stains negative
with this antibody.
